The National Institute of Standards and Technology (NIST) has released a new scientific tool designed to accelerate research into the human gut microbiome, an area of study increasingly linked to numerous serious health conditions. The reference material—consisting of meticulously analyzed human fecal samples—will serve as a standard to help researchers ensure accuracy and consistency across studies and potentially pave the way for new treatments based on microbiome manipulation.

Known as RM 8048, the reference material includes eight vials of human feces, drawn from both vegetarian and omnivore donors. These samples were frozen and suspended in aqueous solution, accompanied by detailed data identifying microbes and biomolecules. NIST developed the product over six years with the goal of helping scientists and biopharmaceutical developers produce reliable results and reproducible experiments when studying the gut microbiome’s role in conditions such as obesity, mental illness, cancer, and diabetes.
Recent progress in the field includes the approval of two drugs derived from fecal matter to treat recurrent C. difficile infection, which causes tens of thousands of deaths annually in the U.S. Fecal transplants have also shown high success rates for treating this infection and are being tested for broader use against diseases ranging from colitis to alcoholic hepatitis. NIST officials say that their new standard will be instrumental in advancing similar treatments by improving data comparability across laboratories and institutions.

The gut microbiome is notoriously complex, composed of trillions of microorganisms and thousands of distinct biological components. This complexity has made it difficult for scientists to draw consistent conclusions using different measurement techniques. NIST’s reference material addresses this by offering a benchmark for laboratories to evaluate their methods, facilitating reproducibility and accuracy. It also enables researchers to determine which analytical approaches best suit various research objectives.
To develop the material, NIST collaborated with a medical services firm to collect stool samples from healthy adult volunteers with varying diets. The samples underwent extensive chemical and genetic analysis to identify more than 150 microbial species and over 150 metabolites. The final product ensures consistency and longevity, with a shelf life of at least five years. NIST hopes that RM 8048 will bring greater standardization to a rapidly growing but fragmented area of scientific inquiry, ultimately helping to advance new diagnostics and microbial therapies.
